Identification and Characterization of a Local Bacterial Strain with High Keratinolytic Activity from Chicken Feathers
نویسنده
چکیده
The present study was aimed at isolating a potential keratinase producing bacteria from chicken feathers collected from poultry waste sites in and around Coimbatore, Tamil Nadu, and optimising the parameters for the enzyme production. The strain was identified as Bacillus licheniformis KMBVP based on 16srRNA gene sequencing. The strain showed high keratinolytic activity when cultured in feather meal medium. Optimising parameters such as pH, temperature, incubation time, carbon and nitrogen sources were also determined. Keratinase production was maximum at temperature 40C, pH 8.0 and on the 7 day of incubation. Feather meal and yeast extract were found to be the optimised carbon and nitrogen sources. The enzyme activity was strongly inhibited by PMSF, suggesting that it belongs to the family of serine proteases with a potential application in feather degradation.

Microbial degradation of keratinous wastes is preferred over physicochemical methods as the latter is costlier and not eco-friendly. Novel habitats are promising for discovery of new microbial strains. Towards discovery of novel keratinolytic bacteria, screening of bacterial strains from a novel limestone habitat in Hundung, Manipur, India was done and a promising isolate, MBRL 575, was found t...

In order to develop a practical method for the decomposition of intact chicken feathers, a moderate thermophile strain, Meiothermus ruber H328, having strong keratinolytic activity, was used in a bio-type garbage-treatment machine working with an acidulocomposting process. The addition of strain H328 cells (15 g) combined with acidulocomposting in the garbage machine resulted in 70% degradation...

A new Streptomyces sp. IF 5 was isolated from the feather dumped soil and found to have a tremendous keratinase activity. The strain enabled the degradation of the chicken feathers very effectively in 60 h. The 16S rRNA sequence of 1474 bp long was submitted to the National centre for Biotechnological information. The keratinolytic activity in the culture medium was 1181 U/ml. The release and a...
